摘要:
目的 检测富含半胱氨酸的酸性分泌蛋白(SPARC)在胃癌组织中的表达情况,探讨SPARC对人胃癌细胞株MGC-803增殖、转移能力的影响以及可能的分子机制。方法 采用实时荧光定量聚合酶链反应(RT-qPCR)检测SPARC在59对胃癌组织及配对癌旁组织中的mRNA表达水平,免疫组织化学检测胃癌组织芯片中蛋白表达水平。利用小干扰RNA(siRNA)技术下调胃癌细胞株MGC-803中SPARC的表达,运用CCK-8、平板单克隆实验、Transwell检测敲低SPARC对细胞的增殖、转移能力的影响。结果 胃癌组织中SPARC的mRNA和蛋白表达水平明显高于癌旁组织。与对照组(MGC-803-siNC)相比,敲低SPARC组(MGC-803-siSPARC)的细胞增殖和转移能力明显减弱。结论 SPARC在胃癌组织中表现为明显的低表达,SPARC蛋白高表达的胃癌患者生存期短,预后差,敲低SPARC抑制MGC-803细胞的增殖和转移能力。

Abstract:
Objective To investigate the
expression level of secreted protein acidic and cysteine rich (SPARC) proteins
in gastric cancer tissues and detect the effect of SPARC on the proliferation
and metastasis in MGC -803 cell lines and its possible mechanism. Method
RT -qPCR was used to detect the mRNA expression of SPARC in 59 pairs of gastric
cancer tissues and paired adjacent tissues, and immunohistochemistry was used
to detect the protein expression of SPARC in TMA. SPARC was down-regulated by
small interfering RNA (siRNA) in gastric cancer cell line MGC-803. CCK-8, plate
cloning assay and Transwell were used to detect the effect of SPARC on cell
proliferation and metastasis. Result The expression levels of SPARC mRNA
and protein in gastric cancer tissues were significantly higher than those in
adjacent tissues. Compared to the control group (MGC-803-siNC), the
proliferation and metastasis ability were significantly decreased in SPARC
-knockdown group (MGC-803-siSPARC). Conclusion SPARC is up-regulated in
gastric cancer tissues, and high expression of ZBTB10 predicts a worse
prognosis in patients with GC. SPARC can promote the proliferation and
metastasis of MGC-803 cells.
