The role of osteoglobin in the maintenance of gastric cancer cell stemness and sensitivity to chemotherapy

Abstract:
Objective To investigate the
effect of osteoglycin (OGN) on the stemness and chemotherapy resistance of
gastric cancer (GC) cells. Method Real-time polymerase chain reaction
(RT-PCR),western-blot,immunohistochemistry
(IHC) combined with The Cancer Genome Atlas (TCGA) database to analyze OGN in
GC cells and tissues expression. CCK-8 and clone formation assay were used to
assess changes in cell proliferation ability,and
apoptosis was measured by flow cytometry. CCK-8 was used to detect the role of OGN
in chemoresistance of GC cells. Western-blot detection of cell stemness
-related molecular markers expression changes. Result The expression of
OGN in GC cells was lower than that in gastric mucosal cells (P<0.05).
Overexpressed OGN decreased the invasive ability of GC cells (P<0.05) and
increased apoptosis (P <0.05). After platinum and fluorouracil treatment,the survival rate of cells decreased and chemotherapy resistance was
alleviated (P<0.05),and OGN induced chemotherapy
resistance . Overexpression of OGN suppressed the expression of stemness
markers. Conclusion OGN is weakexpressed in GC tissues,and overexpressed OGN can inhibit cell stemness and affect
chemotherapy resistance.
