Correlation of EphA2 and Ki-67 proteins in colorectal cancer tissues with prognosis of patients

摘要:
目的 探讨大肠癌组织中Eph受体酪氨酸激酶A2 (Eph receptor tyrosine kinase
A2,EphA2)、Ki-67蛋白在大肠癌组织中的表达与患者预后的相关性。方法 选取2016年7至12月收治的行手术治疗的大肠癌患者115例,使用免疫组化法分别检测癌组织和癌旁组织中EphA2、Ki-67蛋白表达情况,分析两种蛋白表达与肿瘤大小、癌组织分化、Dukes分期、淋巴结转移的相关性,Kaplan-Meier曲线对比癌组织中EphA2、Ki-67蛋白表达与患者生存时间的关系。结果 癌组织中EphA2表达率为89.57%,Ki-67表达率为66.96%,均显著高于癌旁组织的23.78%、29.57%;EphA2和Ki-67蛋白表达与癌组织分化程度、Dukes分期、淋巴结转移均密切相关(P<0.05);随访36个月,Kaplan-Meier曲线显示EphA2、Ki-67蛋白表达阴性患者生存率明显高于表达阳性(P<0.05),Cox分析显示EphA2、Ki-67均是影响大肠癌预后的独立危险因素(P<0.001)。结论 EphA2、Ki-67蛋白高表达可能在大肠癌发病、进展中参与度较高,并且可以预测患者近中期预后状况。

Abstract:
Objective To investigate the correlation
between the expression of Eph receptor tyrosine kinase A2 (EphA2) and Ki-67
protein in colorectal cancer tissues and their prognosis. Methods From
July to December 2016, 115 patients with colorectal cancer undergoing surgery
were treated with immunohistochemistry (SP)to detect the expression of EphA2
and Ki-67 protein in cancer tissues and adjacent tissues. Correlation between
the expression of these proteins and tumor size, cancer tissue differentiation,
Dukes stage, and lymph node metastasis. Kaplan-Meier curve was used to compare
the relationship between EphA2, Ki-67 protein expression and cancer survival
time in cancer tissues. Results The expression rate of EphA2 in cancer
tissues was 89.57%, and the expression rate of Ki-67 was 66.96%, which were
significantly higher than 23.78% and 29.57% of adjacent tissues. [(X2= 45.017,
P<0.001), (X2=32.195, P<0.001)]; EphA2 and Ki-67 protein expression are
closely related to the degree of cancer tissue differentiation, Dukes stage,
lymph node metastasis (P<0.05); KM curve showed that patients with negative
EphA2, Ki-67 protein expression were followed up for 36 months Survival rate
was significantly higher than positive expression (P<0.05). Cox analysis
showed that EphA2 and Ki-67 were independent risk factors affecting the
prognosis of colorectal cancer (P<0.001). Conclusions The high
expression of EphA2 and Ki-67 protein may be involved in the onset and progression
of colorectal cancer, and can predict the prognosis of patients in the near to
mid-term.
