摘要:
目的 探讨长链非编码核糖核酸(LncRNA)胃癌相关转录因子11(STCAT11)与胃癌患者临床特征及胃癌细胞生物学行为的关系。方法 在39份胃癌及其癌旁组织中分别利用实时荧光定量PCR法检测STCAT11表达量,并分析表达量与临床特征的关系。在STCAT11低表达及高表达胃癌细胞株中分别转染表达及干扰质粒,应用克隆形成及细胞计数试剂盒8(CCK-8)实验检测细胞增殖能力,通过Transwell小室实验检测细胞侵袭能力、划痕试验检测细胞迁移能力及裸鼠成瘤实验检测成瘤能力。结果 研究发现与癌旁组织相比,胃癌组织中STCAT11显著低表达,并且表达量与TNM分期、浸润深度及淋巴结转移呈负相关(P均<0.05)。体外和体内实验均证实上调STCAT11表达可限制胃癌细胞的增殖及侵袭能力,下调STCAT11表达则起促进作用(P均<0.05)。结论
LncRNA STCAT11在胃癌组织中表达下调,其表达失调可能参与了胃癌发生、发展的过程,提示STCAT11有可能作为胃癌早期诊断和治疗的分子靶点。

Abstract:
Objective
To investigate the relationship between LncRNA STCAT11 and clinical characteristics
of gastric cancer patients and biological behavior of gastric cancer cells. Methods
Quantitative reverse-transcription polymerase chain reaction (qRT-PCR) was
applied to detect the STCAT11 expression in 39 gastric cancer tissues and
gastric cancer cell lines, while analyzing the relationship between the expression
and the clinical significance. Low and high STCAT11 expression gastric cancer
cell lines were transfected separately with expression and interference
plasmid, then the colony formation and CCK-8 assays were applied to detect the
Cell proliferation, the scratch assay for migration, the Transwell migration
assay for invasion. Detection of tumorigenicity by nude mice tumorigenicity
test. Results The expression of STCAT11 was down-regulated in gastric
cancer tissues compared with the adjacent tissues, the difference was statistically
significant (P<0.05 ). The age and gender are not associated
with STCAT11 expression level(both P>0.05 ), while
positively correlated with TNM stage (P<0.05 )、depth of invasion (P<0.05 )and lymph node metastasis
(P<0.05 ). In vitro and in vivo experiments have
confirmed that up-regulation of STCAT11 expression can inhibit the proliferation
and invasion of gastric cancer cells, while down -regulation of STCAT11 expression plays a promotive role(both P<0.05). Conclusions
The expression of STCAT11 is down-regulated in gastric cancer tissues, and its
imbalance may be involved in the occurrence and development of gastric cancer,
suggesting that STCAT11 may be a molecular target for early diagnosis and
therapy in gastric cancer.
